Human Bone Marrow-Derived CD 34 positive cells are a subpopulation of hematopoietic stem cells, present in the niche. The differentiation potential of these cells is highly investigated forming various blood types. This property makes these cells ideal candidates for studies requiring a large number of cells from either single donor or pooled population, or studies investigating differences of mechanism in cells of donors of various ages, genders, and/or ethnicities. Note: In case your studies demand a more naïve population of cells, then cord blood-derived CD34+ cells population is the better alternative. Yes, it is possible. However, for optimum results, we recommend culturing the thawed cells in the recommended medium before nucleofection. Any further enrichment of the cultured cells is not required; however, the viability may go down to a certain extent.

The CD34-positive cells are isolated from a suitable source of collection, by obtaining a group of mononuclear cells; containing a large population of precursor cells. The CD34+ are then separated with the help of a technique called positive magnetic cell sorting using suitable antibodies.

The Kosheeka is determining the cellular purity of CD34+ with the help of FACS analysis for CD 34 specific expression markers. The Kosheeka makes sure that the expression of these markers is more than or equal to 90%. We are providing the details of the characterization study along with the Certificate of Analysis, of each lot.

Kosheeka’s CD34+ cells should be thawed according to the standard procedure for thawing poietic cells. Following the standard procedure, we typically suggest culturing CD34+ cells using a seeding density of approximately 20,000 to 2,00,000 cells per ml of hematopoietic growth medium (HCGM), with or without serum and supplemented with suitable cytokines. It has to be noted that these cells cannot be expanded beyond 10 population doublings; after which they will get differentiated. We also suggest that if your experiment needs to retrieve the stemness of these cells; then these cells should be maintained in the culture without proliferation and expansion.

We have not analyzed the size of the CD34+ cells, however, the literature is suggesting that when resting these cells are about 8-10 microns in size.

As per the literature support, it can be confirmed that the total population of CD34+ cells in bone marrow mononuclear cell fraction is around 2-3%.

The population of CD34+ cells isolated from different sources differs in their potency for being produced at a different level of tissue differentiation. Since cord blood is formed earlier during fetal development, the cells isolated from this source are comparatively naïve with higher potency of cellular differentiation and trans-differentiation potential; while bone marrow-derived cells are more mature and multipotent in nature, with a capability to differentiate into cells of blood origin only.

The cells will expand and differentiate simultaneously once placed in suitable cultured conditions. You can vary the cytokine cocktail to see if they can be maintained in cell culture without expansion and differentiation.
