Ethically sourced from liver tissues, stellates are the specialized cell types primarily responsible for storing retinoids. However, in response to liver damage, they undergo a transformation, losing their stored retinoids and becoming proliferating fibroblast-like cells. These activated stellate cells deposit collagen matrix, leading to the accumulation of collagen in the liver, a condition known as liver cirrhosis. In order to study stellate cells, they can be isolated from liver tissues; where they are enriched in their population and can be cultivated further in culture. Once in culture, stellate cells immediately differentiate into their activated state, characterized by high expression of alpha-smooth muscle actin (alpha-SMA).At Kosheeka, we are isolating stellate cells from liver tissues and are growing them in culture with the help of specialized media. These cells are mainly characterized for the stellate cell morphology after cryopreservation and thawing. The expression of vimentin is specifically checked to understand the quality of the cells isolated; this protein is primarily assessed with the help of flow analysers. We can also perform immunohistochemical experiments on demand, for the presence of alpha smooth muscle actin, a key marker of activated stellate cells.

Hepatic stellate cells are anchorage dependent cells and can be easily expanded up to 5-6 passage. More number of passages will depend upon the individual’s handing and laboratory conditions.

Several markers are commonly used to identify and characterize hepatic stellate cells. Some of the key markers include desmin, glial fibrillary acidic protein (GFAP), and lipids containing retinoids. However, during activation, the cells mainly express alpha-smooth muscle actin (alpha-SMA) and hence, we are characterizing cells with the said markers.

Knowing when to stop culturing hepatic stellate cells depends mainly upon the specific experimental goals and characteristics of the cells in culture. However, on a generalized note, if the cellular growth is stunted and cells take longer duration to achieve confluence; it is recommended to stop culturing them further. In some cases, you may observe that cells are not attaining their characteristic spindle-shaped morphology due to being in stress or ageing; and that is when you should stop culturing them further.

Finding black, circular spots in your culture can indeed be very concerning and may indicate a potential mycoplasma contamination. You can confirm whether mycoplasma are present in your culture with the help of fluorescent stain and/or polymerase chain reaction. If the mycoplasma are positive, you can immediately discard all the available cultures in your lab, quarantine the space, and reestablish the cultures after thorough cleaning and decontamination of the lab.

These cells can be easily detached from the surface with the help of Trypsin-EDTA. In some cases, you can also use cellular scrapper to detach them from the flasks.
