Dental pulp stem cells are a type of mesenchymal stem cells that have shown great promise in treating stomatognathic disorders. Due to their success at preclinical as well as clinical levels, they are the preferred research tools, especially in vitro research pertaining to maxillofacial and periodontal disorders. The adult dental pulp stem cells (DPSCs) are isolated with ethical consent from the third molars of the adult person; during the extraction of wisdom teeth. During characterization, it was observed that these DPSCs express similar structural and functional markers as that of mesenchymal stem cells, isolated from other well-known sources, like wharton’s jelly, bone marrow, etc. Similar to the MSCs isolated from other sources, DPSCs also exhibit transdifferentiation potential to chondrogenic, adipogenic, and osteogenic lineages. The DPSCs can be expanded up to ten passages to express markers like CD105, CD90, CD73 and are found to be negative for markers like CD34 and CD45. Cells are tested to be negative for mycoplasma, bacteria, yeast and fungi. On demand, cell extract is also tested for HIV-1, Hepatitis C and Hepatitis B.Each lot of cells comes with a detailed certificate of analysis.

Mesenchymal stem cells are preliminarily involved in repairing and maintaining the cellular balance inside human bodies. When tissues get worn out or damaged due to diseased conditions, these cells take charge of the battle and play a lead role by helping to fix things up. These cells secrete various anti-inflammatory cytokines to promote healing. The MSCs isolated from different sources, exhibit the property of a source, from which they are isolated. In xenograft models, the DPSCs are successfully differentiated into mineralized tissue, extracellular matrices like dentin, dental pulp and periodontal ligaments.
A dental pulp is obtained from individuals, undergoing wisdom tooth extraction with the help of informed consent. After extraction, the pulp is carefully transported to the laboratory in an appropriate antibiotic solution. Prior to the extraction of DPSCs from a tissue, a thorough washing of the tissue is conducted to eradicate the chances of tissue contamination. The isolation of dental pulp stem cells is achieved using explant techniques.

Karyotyping analysis of extracted dental pulp stem cells is performed to examine genetic makeup of cells. This is mainly with respect to the therapeutic application of these stem cells.
Screening for infectious markers like HIV, HCV, HBV is conducted at passage 1.
At passage 1, surface marker analysis of stem cells is done via BD FACSCalibur Flow Cytometry. For assessments, positive markers like CD29, CD73 and CD105 are used; while negative markers used are CD34 and CD45.
The differentiation potential of isolated DPSCs is assessed using suitable differentiation kits for adipogenic, chondrogenic as well as osteogenic potential.

Mesenchymal stem cells are robust and thrive under standard culture conditions, typically incubating at 37 degrees Celsius with 5% carbon dioxide. Proper handling requires familiarity with aseptic techniques and general cell culture procedures, such as cell passaging, cryopreservation, and revival. These cells can be effectively cultivated in regular media like DMEM/F12 supplemented with 10% fetal bovine serum.
For long-term storage, cells are preserved in liquid nitrogen using a medium consisting of 90% fetal bovine serum and 10% dimethyl sulfoxide (DMSO).
