Due to their distinct degrees of potency, these cells are being extensively studied for their applications in regenerative medicines and tissue engineering. Mesenchymal stem cells are multipotent adult stem cells, isolated from the stroma of adipose tissue and are of mesenchyme origin. They are multipotent in nature due to their ability to differentiate into cells of multiple origins including bone, cartilage, connective tissue, neurons, etc. They have fibroblast-like morphology and are adherent to culture vessels.
On average, the doubling time of preadipocytes is calculated to be between 48-54 hrs, depending upon culture condition and growth supplement. However, it is also very important to keep in mind that the rate of replication of preadipocytes, varies considerably after handling.
The cumulative population doublings are calculated using the seeding density and harvesting density from each type of culture vessel. The cell doubling time is calculated using the formulaDoubling Time = Duration * log (2)/ log [Final concentration/Initial Concentration]
The mesenchymal stem cells isolated from visceral adipose tissue possess high proliferation capacity and hence can be easily expanded up to 7-8 passages; post which, cells start undergoing morphological and chromosomal changes. We are shipping cells generally at P2, however, we can also deliver cells at the early passage, as per the client’s choice.
Cells are generally shipped in the adherent form in a T25 flask at room temperature, within India. However, for delivering cells outside India, cells are suspended in a specific growth medium at a refrigerated temperature. For expanding these cells in culture, we recommend specific media along with a combination of growth factors, depending upon the applications. If cells are to be used for preclinical analysis or therapeutic applications, then it is recommended to grow them in serum-free media to avoid zoonotic infections.
We are using TrypLe for cellular detachment. Interestingly, mesenchymal stem cells are highly versatile and sturdy in nature, and hence, any commonly used detachment enzyme including Trypsin-EDTA, Dispase, etc. can be used for detachment.
While our certificate of analysis has detailed information on donor demographics, including source, donor’s age, sex, and BMI; we are also following regulatory guidelines to prevent pathogenic transmission. Before collecting samples from the donor, each donor is tested for infectious pathogens specified by the regulatory guidelines like HIV-1 & 2, HCV, HBsAg, HTLV-1 & 2, CMV, etc. The details shall be specified in the certificate of analysis
Each batch is quality checked for mycoplasma, sterility, and viability. We are also assessing mesenchymal stem cells for their markers that are specific to them, such as CD105, CD90, etc.
